no you can start with the ones you have already, if you wish. You have to cut and paste every line anyway. It may be even easier to have open only the few characters you have so far, instead of the 20 later on.
I would start by opening your narrator for act 1, and then all the files you have already for act 1.
Then let your narrator run while reading with the text. As soon as there is a line by someone where you already have the file, go to that recording, cut out the line you need and paste it into the correct slot. At the end, you need to have all the lines in the narrator file and no more lines in the other files. Then you are done with a first pass. Save it maybe under a different name to keep track.
As soon as a new reader comes in, you can cut and paste those files as well, and slowly you will have a whole filled act.
In a second pass then, when all the lines are in, I would relisten to the whole act and maybe adjust a volume, or make more or less silence between quotes, so that it's not too fast and not too slow. Things like that.
Also merging the group voices you can only do when you have all the files for that group in, so that is probably the last thing you copy into the masterfile.
